卡介苗对膀胱癌细胞Toll信号通路相关膜分子基因表达影响
Expression of Toll signal pathway-associated membrane molecule in bladder cancer cell line stimulated by Bacillus Calmette-Guerin
目的 探讨卡介苗对膀胱癌细胞株T739 Toll信号传导相关膜分子TLR2、TLR4、CD14和MD-2 mRNA基因表达影响。方法 采用SYBR Green嵌合荧光定量Real Time-PCR,观察BCG(0.25mg/ml)刺激T739后不同时间点各个mRNA相对表达量(Ratio),设LPS和空白组作对照。结果 BCG组TLR2、CD14和MD-2 mRNA在BCG刺激后表达增加,Max.Ratio分别为5.3(2h), 2.6 (4h), 7.4(2h),TLR4无明显变化;LPS组CD14和TLR4表达增加,Max.Ratio分别为2.1(1h),3.2(6h),TLR2和MD2无明显变化。空白组上述基因各时间点Ratio无明显变化。结论 BCG和LPS作用T739的受体不同,推测卡介苗可经TLR2激活膀胱上皮细胞Toll信号通路。
Purpose To explore the expression of Toll signal pathway-associated membrane molecules in mouse bladder cancer cell line T739 stimulated by BCG. Methods BCG was added to culture for stimulation at final concentration of 0.25mg/ml; The levels of TLR2、TLR4、CD14,MD-2 mRNA were measured by relative quantitative RT-PCR using SYBR Green fluorochrome at variable time points. As control, we arranged LPS group and blank group. Results The levels of TLR2,CD14 and MD-2 mRNA were increased by the stimulation of BCG, and the Max.Ratio of them were 5.3 at 2 h , 2.6 at 4 h, 7.4 at 2 h after BCG stimulation respectively; however change of TLR4 mRNA has no significance. The levels of TLR4 and CD14 mRNA were increased by the stimulation of LPS, but changes of TLR2 and MD-2 mRNA have no significance. Change of all mRNA levels has no significance in blank group. Conclusions BCG has different receptor compared with LPS, and Toll signal pathway may be actived by BCG through TLR2 in Urothelial Cell.
