目的:探讨复方浙贝颗粒联合阿霉素对P388移植瘤的p53基因表达影响。方法:将小鼠淋巴细胞白血病细胞系P388细胞接种于昆明小鼠腋前皮下构建小鼠移植瘤模型,予复方浙贝颗粒联合不同剂量的阿霉素灌胃/和腹腔注射治疗。治疗14d后处死小鼠,剥离肿瘤,荧光定量聚合酶链式反应法检测各组移植瘤组织p53基因表达,2-ΔΔCt法计算各组p53基因的相对定量。结果:各组移植瘤中p53基因相对表达的2-ΔΔCt值比较,差异无统计学意义(F=0.56,P=0.7557)。其中,CZBG中剂量联合ADM大剂量组p53基因相对表达量较高,而CZBG中剂量联合ADM小剂量组p53基因相对表达量较低。结论:CZBG与ADM联合使用似乎能够上调P388移植瘤中p53抑癌基因的表达。

Objective Studying the impact on p53 gene expression which is caused by compound zhe-bei granule and adriamycin in the P388 xenograft tumor. Methods Establishing tumor xenografts model by injecting the mouse lymphocytic leukemia cells (P388) in the subcutis of anterior axillary of Kunming mice, then treating with CZBG by intragastric administration and different doses of adriamycin by intraperitoneal injection (i.p.). After the end of the experiment, striping tumor completely, detecting the expression of p53 gene in xenograft tumor of each group by fluorescence quantitative polymerase chain reaction, and computing the relative quantification of p53 gene of each group using 2-ΔΔCt method. Results Comparing the 2-ΔΔCt values of p53 gene relative expression of each group, no statistical significance is found (F=0.56, P=0.7557). And the relative expression value of p53 gene of CZBG Joint high-dose ADM group is higher, while the relative expression value of CZBG Joint low-dose ADM group is lower. Conclusion Combined use of CZBG and ADM seems to be able to raise the expression of p53 tumor suppressor gene in the P388 xenograft tumor.
