Study on the Inhibition Effect and Mechanism of Breast Cancer Cell Line MCF-7 by Ginseng Rust Rot

目的研究锈腐病人参对MCF-7细胞增殖的抑制及作用机制。方法使用SEM和AFM研究MCF-7细胞的形貌及力学特性的变化,对MCF-7细胞的克隆形成能力进行检测,蛋白免疫印迹法检测MCF-7细胞中β-actin蛋白的表达水平。结果与对照组相比,随着药物浓度的增加,细胞间隙变大,细胞平均高度值增大,表面粘附力和杨氏模量均呈现下降趋势。杨氏模量值变化显著。克隆形成实验显示MCF-7细胞的克隆能力被显著抑制,WB实验结果显示细胞内β-actin蛋白表达水平显著降低。结论锈腐病人参可抑制MCF-7细胞增殖,可能与细胞内β-actin蛋白表达水平下降有关。

Objective The purpose of this article is to study the inhibitory effect of ginseng rust rot on the proliferation of MCF-7 breast cancer cells and its mechanism. Methods The cell morphology and mechanical characteristics of the MCF-7 cells treated with different concentrations of ginseng rust rot were investigated by SEM (scanning electron microscopy) and AFM (atomic force microscopy), respectively. The plate clone formation experiment was used to test the clonal formation abilities in vitro. The expression of β-actin in MCF-7 cells was detected by Western blot. Results Compared with the control cells, both the morphology and mechanical properties of MCF-7 cells changed greatly after treated with the extract of ginseng rust rot. These changes were dose dependent. With the increased concentration of ginseng rust rot extract, more proteins were observed on the surface of MCF-7 cells. The distance between cells became larger. The cell average height gradually increased, while the average adhesion and the average Young\'s modulus declined, especially for the decrease of 2.37 kPa in the average Young\'s modulus. In addition, the cloning ability of MCF-7 cells was significantly inhibited, and the expression of β-actin protein was also significantly reduced. Conclusion Ginseng rust rot can inhibit the MCF-7 cell proliferation, and it may be related to the decline of intracellular β-actin.?
