新烟碱类农药宽谱单克隆抗体制备与性能鉴定
Production and characterization of the broad-specific monoclonal antibodies against neonicotinoid pesticides
本文采用同源竞争ELISA模式,以吡虫啉和啶虫脒两种农药竞争筛选,获得了新烟碱类农药的宽谱单抗。采用间接竞争ELISA法测定其灵敏度和特异性,结果表明该单抗在吡虫啉-OVA同源包被下获得最优灵敏度,其中对吡虫啉、氯噻啉、噻虫胺三种农药的亲和力较好,IC50分别为0.27、0.24、0.61 ng/mL;与啶虫脒、噻虫啉、烯啶虫胺三种农药的亲和力较前三种略差, IC50分别为2.70、0.95、3.77 ng/mL。获得的此株宽谱抗体可用于建立吡虫啉、氯噻啉、噻虫胺、啶虫脒、噻虫啉和烯啶虫胺这六种新烟碱类农药的定性或半定量的多残留免疫快速检测方法。
Homologous ELISA was introduced into the screening of hybridomas with the competition of imidacloprid and acetamiprid for the development of broad-specific monoclonal antibodies (MAbs) against neonicotinoid pesticides. By indirect competitive ELISA method (ic-ELISA) to measure its sensitivity and specificity, the results show that the MAb combined with imidacloprid -OVA could be the best recognizability. It can respectively recognize imidacloprid, imidaclothiz and clothianidin, with IC50 values of 0.27、0.24、0.61 ng/mL. Besides, it can also recognize acetamiprid, thiacloprid and nitepyram, with IC50 values of 2.70、0.95、3.77 ng/mL. The obtained broad-specific MAbs can be used for the rapid qualitative or semi-quantitative multi-analyte immunoassay detection of the six kinds of neonicotinoid pesticides.
