蕨菜多糖的分析

本文研究了采用DEAE-Sepharose Fast Flow 和Sephacryl S-400 HR 柱层析纯化后的蕨菜多糖的结构和单糖组成。采用紫外光谱和傅立叶转换红外光谱法对蕨菜多糖进行定性分析;采用气相色谱法测定蕨菜多糖中单糖的种类和构成比例;采用凝胶色谱法测定蕨菜多糖的分子量。结果表明,蕨菜多糖具有多糖特征性的紫外和红外吸收峰; 根据用不同分子量的标准葡聚糖做出标准曲线,测定出纯化后的蕨菜多糖PLP分子量(Mw)为458000Da;气相分析结果表明蕨菜多糖中葡萄糖含量最高(相对含量58.1%),其次是由半乳糖、甘露糖、鼠李糖、阿拉伯糖(18.7%, 6.8%, 10.2% 和6.1%)等5种单糖组成。
In this paper, monosaccharide composition and the structure of a water-soluble polysaccharide, obtained from Pteridium aquilinum , was studied by ultraviolet(UV) , Fourier transform infrared spectroscopy (FT-IR) and gas chromatography(GC). The water-soluble polysaccharide has been fractionated by DEAE–Sepharose Fast Flow column chromatography and purified by Sephacryl S-400 HR column chromatography. The Fourier-transform infrared spectra (FT-IR) of the purified polysaccharide (PLP) revealed typical characteristics of polysaccharides. The weight-average molecular weight (Mw) of PLP was calculated to be 458000Da, according to the calibration curve with standard dextrans. Gas chromatography (GC) traces of the polysaccharide hydrolyzates indicated that glucose (relative mass 58.1%) was the predominant monosaccharide of PLP with low levels of galactose, mannose, rhamnose and arabinose (relative mass 18.7%, 6.8%, 10.2% and 6.1%).
