溶菌酶水解壳聚糖的机理研究

本文运用粘度法、还原端基法等方法,研究了溶菌酶水解壳聚糖过程中温度、pH值、反应时间、底物的浓度和脱乙酰度等对反应速度的影响。结果表明:溶菌酶能有效降解壳聚糖,其作用的最适温度为45℃,最适pH值为5.4,米氏常数为Km=0.055mg/ml,Vmax=18.1476U/ml,底物的专一性较强,随着底物脱乙酰度的增加反应速率下降。在最适条件下水解30min,底物粘度迅速下降80%,初步分析判断溶菌酶以内切方式作用于壳聚糖。

he effect of reaction temperature, pH, time, substrate concentration, the deacetylation degree of chitosan (DDA) on the chitosan degradation by lysozyme were investigated. The result showed that the optimal condition for depolymerization of chitosan with lysozyme were at pH 5.4 and 45℃,respectively. The data revealed that the km value was 0.055mg•ml-1and Vmax was 18.1476U•ml-1. The relative activity decreased when degrees of deacetylation increased .The best substrate was 78% deacetylated chitosan.
