以原核表达的鸡白细胞介素-2受体α链(chIL-2Rα)重组蛋白为免疫原,免疫BALB/c小鼠,利用细胞融合技术获得了7株稳定分泌抗chIL-2Rα单克隆抗体(mAbs)杂交瘤细胞(4H5、6C9、6A1、5E1、1G1、6C2、1E1),单抗轻链均为kappa链,重链为IgG1(1E1为IgG2b)。Western blot和免疫细胞化学检测显示,7株单抗均能与特异识别原核和真核细胞表达的chIL-2Rα蛋白。

In this paper, seven hybridomas secreting monoclonal antibody (mAbs) of 4H5, 6C9, 6A1, 5E1, 1G1, 6C2 and 1E1 sepcific for chicken interleukin-2 receptor α chain (chIL-2Rα) were established by immunizing mice with the recombinant protein, the Ig isotypes of which were kappa for light chian and IgG1 for heavy chain except for 1E1 (IgG2b). The results of Western blot and immunocytochemistry showed that all these mAbs could recognized the recombinant chIL-2Rα protein expressed in the prokaryotic and eukarytic cells.
