酸枣仁汤对老年阴血亏虚型失眠证候模型大鼠学习记忆及脑内自由基、NOS的影响
Effects of SZRT on learning memory and levels of free radicaI and NOS in old model rats brain with Yin and blood-deficiency(阴血亏虚) and sleep deprivation
目的:探讨酸枣仁汤对老年阴血亏虚失眠证候模型大鼠学习记忆及脑组织MDA含量及SOD、一氧化氮合酶(NOS)活性的影响。 方法:用D-半乳糖致衰老、环磷酰胺及氢化可的松致阴血亏虚,及自制改良多平台法剥夺睡眠,制作老年阴血亏虚型失眠证候大鼠复合模型,观察大鼠学习记忆及大脑皮质及海马部位MDA含量及SOD、一氧化氮合酶(NOS)活性的变化,并考察酸枣仁汤对这些指标的影响。结果:与环境对照组比较,证候模型组睡眠剥夺48h后,大鼠学习记忆能力显著下降,脑组织MDA含量和SOD、NOS活性均升高;经酸枣仁汤干预后大鼠学习记忆能力得以明显改善,脑内各指标明显下降(P<0.01, P<0.05),并趋于正常范围。结论:酸枣仁汤对老年阴血亏虚失眠证候模型大鼠的学习记忆能力障碍有改善作用,且酸枣仁汤大剂量组作用略优于小剂量组,其机理之一可能与减轻睡眠剥夺中自由基和NO对神经细胞的损伤有关。
Object:To observe the effects of SZRT on learning memory and the levels of malondialdehyde(MDA) and the activities of supemxide dismutase(S0D) and nitric oxide synthase (NOS) in old model rats brain with Yin and blood-deficiency(阴血亏虚) and sleep deprivation(SD). Methods:levels of learning memory and MDA, and activities of SOD and NOS in mice brain were assayed after SD induced by maded-self multiple platform method(MMPM),senescence induced by D-galactose,and Yin and blood-deficiency induced by cyclophosphamide and Hydrocortisonum. Results:After 48 SD,levels of learning memory and MDA, and activities of SOD and NO in mice brain markedly increased in comparison to the control group.However,the observation of the groups treated with SZRT were significantly decreased(P<0.01, P<0.05),and showed a tendency in returning to normal leve1.Conclusion:SZRT can improve the disability of learning and memory of the pathogeny rats model, and its one of mechanis maybe involve the reduction of nevours cell damage with free radical and NO.What’s more,the high dose of SZRT is better than that of the low one.
