目的:明确电门控质子通道蛋白Hvl对乳腺癌细胞迁移及侵袭能力的影响。方法:检测电压门控质子通道蛋白Hv1在不同转移能力的人乳腺癌细胞系中的表达,利用小RNA干扰(siRNA)技术下调Hv1在乳腺癌MDA-MB-231细胞中的表达,通过细胞划痕和体外侵袭实验方法观察Hvl对乳腺癌细胞迁移和侵袭的影响并初步探讨相关分子机制。结果:Hv1在高转移的乳腺癌细胞MDA-MB-231中表达较高;Hvl基因的siRNA干扰片段能够抑制Hvl基因及蛋白的表达;细胞划痕和体外侵袭实验表明Hvl降表达的MDA-MB-231细胞迁移和侵袭能力较弱;明胶酶谱和免疫印迹实验证明下调Hvl基因在MDA-MB-231细胞中的表达明显抑制了MMP-2的活性。结论:电压门控质子通道蛋白Hvl能够促进乳腺癌细胞迁移及侵袭。

Objective: Clarify the effect of Voltage-gated proton channel(Hv1) on the migration and invasion of breast cancer cells. Methods: The protein expression of Hv1were detected in human breast cancer cell lines with different metastatic ability. SiRNA technique was used to down-regulate the expression of Hvl in breast cancer MDA-MB-231 cells, scratch and matrigel invasion methods were used to observe the effect of Hvl on migration and invasion of breast cancer cells, and the relevant molecular mechanism were explored. Results: Hv1 is highly expressed in the highly metastatic breast cancer MDA-MB-231 cells. Hvl was expressed higher in the MDA-MB-231 cells with high metastatic ability. The SiRNA sequence target at Hvl could inhibit the expression of Hvl. Scratch and matrigel invasion experiments showed that the migration and invasion of MDA-MB-231 cells were significantly attenuated when Hv1 was knocked down by siRNA targeting to Hv1 and the result of Zymography experiment of matrix metalloproteinase indicated that the enzyme activities of MMP-2 was markedly decreased. Conclusion: Hv1 promotes the migration and invasion ability of breast cancer cells.
