家蚕蛹油的提取工艺优化及其脂肪酸的性状和组分分析
Optimization of the extraction parameters,fatty acid character and composition analysis of bombyx mori chrsalis oil
本研究利用蚕蛹为原料,研究了家蚕蛹油的提取工艺。以浸提温度、浸提时间、溶剂用量、振荡速率为因素,出油率为考察指标,采用单因素实验和正交试验优化蚕蛹油的石油醚浸提工艺。结果表明,各因素的影响主次顺序为:溶剂用量>浸出时间>振荡速率>浸提温度,最佳提取工艺参数为:浸出温度为40℃,溶剂用量为4mL/g,浸出时间3h,振荡速率为160rpm。该条件下蚕蛹油的出油率达28.13%。采用GC-MS分析了蚕蛹油的脂肪酸组分,所制备的蚕蛹油富含不饱和脂肪酸,含量高达68.34%。其中,主要不饱和脂肪酸为油酸、和α-亚麻酸,含量分别为35.44%和32.18%。另外,所含饱和脂肪酸主要为棕榈酸和硬脂酸,含量分别为31.36%和0.63%。
In this research, the silkworm pupaes were used to extracting pupal oil.Single factor experiments and orthogonal array design were adopted to optimize the parameters of silkworm pupal oil extraction by petroleum ether. The effects of independent parameters, including temperature, extraction time, solvent volume and oscillation rate on the yield of oil were studied. The statistical analysis show that the solvent volume was the most significant variable followed by extraction time, oscillation rate and temperature. The optimal extraction condition for oil yield was temperature 38℃, extraction time 2 hours, solvent volume 4mL/g and oscillation rate 160rpm. Under this condition, the extracting rate of pupal oil was measured about 28.13%. Fatty acid composition of pupal oil obtained was analyzed by GC-MS . The silkworm pupae oil was rich in unsaturated fatty acids which accounted for more than 68.34% of the total fatty acids. The main unsaturated fatty acids are oleic acid (35.44%),α-linolenic acid (32.18%), The main saturated acid in the silkworm pupal oil was palmitic acid (31.36%), followed by stearic acid (0.63%).
