匹罗卡品致痫小鼠海马EphA4蛋白动态表达变化

目的 动态观察匹罗卡品致痫小鼠海马EphA4蛋白的时空表达变化,探讨其在癫痫发生发展中的作用。方法 SPF级C57BL/6 雄性小鼠48只,随机分成正常对照组和致痫后7天、14天和28天组。匹罗卡品腹腔注射制备癫痫模型,应用免疫组化技术检测致痫后各时间点EphA4蛋白表达变化。结果 EphA4蛋白主要分布于齿状回颗粒细胞层、颗粒细胞下层和门区,CA1和CA3区锥体细胞层。模型组海马齿状回、CA3和CA1区EphA4蛋白表达逐渐升高,28天达到最高峰,与对照组相比有显著性差异(p < 0.001)。不同时间点的对照组之间表达无明显差异。结论 癫痫持续状态诱导EphA4表达变化,可能参与了癫痫后异常神经元网络形成过程。
In this paper we studied the dynamic expression pattern of EphA4 protein in the hippocampus of a mouse model of temporal lobe epilepsy induced by pilocarpine. Forty-eight SPF C57BL / 6 mice were randomly divided into control group and status epilepticus (SE) group. The SE group was subdivided into three groups according to the duration of the behavioural features, respectively in the acute period (7 days), the latent period (14 days) and the chronic period (28 days). SE was induced by intraperitoneal injection of pilocarpine. Immunohistochemical staining was used to detect the expression of EphA4 protein in the hippocampus at different time points after SE. EphA4 protein was mainly distributed in the granule cell layer of dentate gyrus, subgranular zone, hilus, pyramidal cell layer of CA1 and CA3 areas. The expression EphA4 protein gradually increased and peaked at 28 days post SE. There was significant difference between SE group and control group at 28 days (p < 0.001). No significant difference between the control groups. The upregulation of EphA4 protein expression indicated that EphA4 may be involved in the formation of abnormal neuronal network.
