猪链球菌9型5'核苷酸酶克隆表达及特性分析

前期研究表明5'核苷酸酶(NT)是猪链球菌9型(SS9)的毒力因子,但对其生物学功能了解有限。序列分析发现NT蛋白中包含5'核苷酸酶活性域,本研究利用磷酸根离子检测试剂盒对SS9野生株GZ0565、NT缺失株Δnt和NT互补株C-nt的酶活性进行测定。结果显示,在与0.2 M AMP作用时,Δnt分解AMP的能力显著低于GZ0565和C-nt。同源性分析发现NT在不同血清型的19株链球菌中都存在,较为保守。另外,以SS9菌株GZ0565基因组DNA为模板扩增NT C端非跨膜区1026bp大小的基因片段,克隆到PMD19-T载体后,插入表达载体PET32a,将重组载体转化入大肠杆菌BL21(DE3),在IPTG的诱导下表达了大小约57 kDa左右的融合蛋白。将表达的蛋白纯化后免疫新西兰大白兔,制备多克隆抗体,三免后血清效价达到1:12800。提取SS9 GZ0565细菌细胞壁蛋白,与兔抗NT重组蛋白血清进行免疫印迹,表明该蛋白位于细菌表面;同时该蛋白可被SS9猪康复血清识别,表明NT具有免疫原性,且在SS9感染过程中表达。上述结果表明NT是一免疫原性蛋白,且在猪链球菌中较为保守,是较为理想亚单位疫苗候选;同时NT具有降解AMP能力,为进一步了解其在SS9致病中的作用奠定基础。
Previous studies showed that 5'-nucleotidase is a virulence-associated factor for Streptococcus suis serotype 9 (SS9), but its biological function is not clear. Since NT contains a 5'-nucleotidase domain, we tested whether it could hydrolyze AMP. The hydrolysis activity of the Δnt mutant was significantly reduced compared with the wild-type strain GZ0565 and NT complemented strain C-nt in the presence of 0.2 M AMP. Homology analysis revealed that NT is conserved in 19 different serotypes SS strains. NT fragment from C-terminal (1026bp) was amplified from SS9 strain GZ0565 genomic DNA and cloned into the PMD19-T vector, then inserted into expression vector pET32a. The recombinant plasmid was transformed into E.coli BL21(DE3). After induction by IPTG, a recombinant fusion protein about 57kDa was detected. A rabbit was immuned with recombinant NT protein (rtNT). After the third immunization, the titer of the serum is 1:12800. The bacterial cell wall proteins of the GZ0565 and the Δnt mutant were extracted and probed with rabbit anti-rtNT serum. An apparent band was detected in the cell wall protein of the GZ0565 strain, while no specific band was observed with the same serum when blotting Δnt cell wall proteins. In addition, NT was recognized by anti-SS9 convalescent sera from pigs. Taken together, we confirmed that NT is conserved in SS strains, a immunogenic protein expressed on the bacterial surface, and a promissing vaccine candidate. It also can hydrolyze AMP, which contributes to understanding its role in SS9 pathogenesis.
