目的:基于以往溶血磷脂酸(LPA)受体在出生后早期大鼠心脏的高表达的研究基础探讨溶血磷脂酸(LPA)信号在心肌再生过程中的调节作用。方法:利用新生大鼠心尖切除模型,将动物分为假手术组、心尖切除组、心尖切除+LPA组、心尖切除+LPA1/3受体抑制剂组,HE和MASSON染色评价心肌修复,免疫荧光观察心肌细胞增殖,超声评价心功能。结果:成功建立大鼠心尖切除模型,切除后21天大鼠心肌不能完全由心肌组织进行修复,但心功能与假手术组无显著差异。LPA可以明显加速切除后心肌修复,并且可以促进增殖心肌细胞数量,而LPA受体抑制剂则可以完全阻断LPA的促增殖作用。结论:LPA-LPA1/3受体可以通过促进心肌细胞增殖加速心尖切除后心肌修复,本研究从内源性脂类信号分子角度为哺乳动物心肌再生调节提供了新线索。

Objective: Our previous study demonstrated that cardiac expression of lysophosphatidic acid (LPA) receptors is much higher during early postnatal period. in this study, we investigated the role of LPA signaling in mammalian heart renegeration. Methods and results: we surgically resected the left ventricular apex from 1-day-old rats. Serial histological analysis revealed progressive regeneration of the apex. At 7 day post-resection (dpr), the resection plane was characterized by a robust inflammatory response in control group while replaced by cardiomyocytes in LPA-injected group detected by morphological analysis. By 21 dpr, the most part of apical defect was replaced by cardiomyocytes and the small part was fibrosis, as detected by morphological analysis and masson staining. Left ventricular ejection fraction was assessed by echocardiography 7 and 21 days after apical resection, the results showed that the newly formed apex had normal systolic function, similar to sham operated hearts. What is more, we found LPA promoted proliferation of cardiomyocytes at 2-14 dpr. And this proliferation of LPA was completely inhibited after intraperitoneal injection of Ki16425 (LPA1/LPA3 inhibitor). Conclusions: LPA signaling promotes the restoration of the heart of neonatal rats responds to injury by cardiomyocyte proliferation, suggesting its therapeutic potential for enhancing immature heart growth in the conditions of congenital heart disease.
