目的:研究利拉鲁肽对糖尿病牙周炎体外模型的治疗作用,并初步探讨其作用机制。 方法:应用倒置显微镜、免疫荧光法、茜素红染色鉴定人牙周膜细胞。CCK-8实验筛选建立糖尿病牙周炎体外模型的最佳药物浓度及时间。qRT-PCR和ELISA检测利拉鲁肽对糖尿病牙周炎模型中IL-6和TNF-α mRNA和蛋白的影响。通过qRT-PCR、ELISA和Western blot探究利拉鲁肽影响糖尿病牙周炎的作用机制。 结果:100 μg/mL AGEs,1 μg/mL Pg-LPS处理人牙周膜细胞24 h建立了糖尿病牙周炎体外模型。糖尿病牙周炎模型中IL-6、TNF-α、JAK2、STAT3表达上调,利拉鲁肽处理可逆转该结果。加入香豆霉素C-A1(JAK2激动剂)后,JAK2、STAT3、IL-6、TNF-α表达上调。 结论:利拉鲁肽通过抑制JAK2/STAT3信号通路对糖尿病牙周炎产生治疗作用。利拉鲁肽有望成为糖尿病牙周炎的新兴治疗药物。?????

Objective: to study the therapeutic effect of lillarutide on diabetic periodontitis in vitro and to explore its mechanism. Methods: human periodontal membrane cells were identified by inverted microscope, immunofluorescence and alizarin red staining. CCK-8 experiment screened the optimal drug concentration and time to establish the in vitro model of diabetic periodontitis. qRT-PCR and ELISA were used to detect the effect of liraglutide on IL-6 and TNF-α mRNA and protein in diabetic periodontitis model. qRT-PCR, ELISA and Western blot were used to investigate the mechanism of the effect of liraglutide on diabetic periodontitis. Results: human periodontal membrane cells were treated with 100 μg/mL AGEs and 1 μg/mL Pg-LPS for 24 h to establish an in vitro model of diabetic periodontitis. The expressions of IL-6, TNF-α, JAK2 and STAT3 were up-regulated in diabetic periodontitis model. JAK2, STAT3,IL-6 and TNF-α expression were upregulated after the addition of coumermycin A1 C-A1 (JAK2 agonist). Conclusion: Liraglutide has therapeutic effect on diabetic periodontitis by inhibiting JAK2 and STAT3 signaling pathways. Liraglutide is expected to be a new treatment for diabetic periodontitis.
