目的:研究记忆性CD4T细胞(Tm)在I型糖尿病(T1D)小鼠发病过程中的作用。方法:利用血糖仪测定血糖,按血糖高低分为血糖正常、轻微发病小鼠、严重发病小鼠。取小鼠外周血、脾脏和胰腺,采用细胞表面染色流式细胞仪检测CD4Tm细胞的比例变化;分离纯化脾脏CD4Tm进行CFSE染色细胞增殖检测;组织切片检测胰腺细胞浸润变化。结果:随T1D小鼠发病加重,血液中CD4Tm比例增加;脾脏细胞CD4Tm比例无显著变化;胰腺组织中CD4Tm比例显著增加。脾脏CD4Tm细胞增殖试验利用抗小鼠CD3抗体刺激各组CD4Tm细胞,增殖情况无明显差别;利用胰岛素多肽刺激CD4Tm细胞,随发病加重,细胞增殖显著增多。组织切片表明胰腺淋巴细胞浸润与发病严重程度正相关。

Objective: Objective: In this paper, we aimed to study the function of CD4 memory T cells in the progress of T1D mice. Methods: The mice were confirmed T1D by blood glucose checked by blood sugar machine. The blood, spleens and pancrease were prepared for CD4 Tm test by immunostaining and FACS; The CD4 Tm from spleens were seperated and purified by CD4 Tm isolation kit and proliferation assay after CFSE staining was performed; The pancrease section were made for infiltrated lymphocytes testing. Results: Following the process of T1D, the percentage of CD4 Tm cells in blood increased significantly; There was no differences for the percentage of CD4 Tm cells in spleens; the percentage of CD4 Tm cells in pancrease increased significantly. The purified CD4 Tm cells from spleens proliferated with the stimulation of insulin peptide, but no differences with the stimulation of anti-CD3 mAb. Following the process of T1D, the infiltrated lymphocytes in pancrease increased in the pancrease sections.
