斜纹夜蛾核型多角体病毒Ⅱ型(SpltNPVⅡ)orf29基因的研究

纹夜蛾核型多角体病毒Ⅱ(Spodoptera litura nucleopolyhedrovirus Ⅱ,SpltNPVⅡ)是一种毒力强繁殖率高的新型病毒,有146个开放阅读框(Open reading frames,ORF)。其中orf29基因是一个新型的未知基因,编码211个氨基酸。生物信息学结果表明ORF29没有明显的跨膜区域,不存在信号肽,与SeMNPV-ORF29同源性最高,并属于DUF1251超家族。本研究通过荧光素酶报告基因检测系统分析orf29基因启动子活性,结果显示该基因是个晚期基因;通过荧光定量PCR和蛋白免疫印迹(Western Blot),分析SpltNPVⅡ病毒感染细胞后orf29基因的转录时相以及GP29的表达时相,结果均证实orf29在病毒感染后的18 h开始转录表达,是个晚期基因。免疫细胞化学分析对GP29进行细胞定位,结果显示GP29大量存在于细胞质中,少量存在于细胞核内。
Spodoptera litura nucleopolyhedrovirusⅡ(SpltNPVⅡ)which has 146 ORFs was a new kind of baculovirus with host specificity and strong pest-killing abilities. And orf29 was a unique and new unknown gene which encoded 211 Aa and had never been reported. Bioinformations suggested ORF29 belonged to DU1251 superfamily, didn't have transmembrane domain and signal peptide and had the highest similarity with SeMNPV-ORF29. In this paper, Luciferase reporter gene detecting system was used to analyse promoter activity of the orf29's leader sequence 29P. The result showed 29P was a late promoter and the orf29 was assumed as a late gene. orf29's transcription phase was determined by realtime-PCR. Western blot was used to detected expression phase of orf29. It was confirmed that the transcription and expression of gene orf29 had started from 18 hours later after the virus infected the spli cells until the cells died. Finally, we carried out cellular localization about orf29 in spli cell, and the result of the immunocytochemistry suggested that GP29 exsited in cytoplasm more than in cell nucleus.
