轴突导向因子Slit是一组分泌型的蛋白,主要在神经系统的发育过程中参与调控轴突定向,分支及神经元的迁移等过程。近年来发现Slit也参与肿瘤生长、血管新生及侵袭转移的调控。人成纤维肉瘤HT1080细胞是来源于间叶组织的,高转移性的恶性肿瘤。本研究检测了slit3在HT1080细胞中的表达情况,并对slit3对HT1080细胞的增殖和侵袭转移进行了研究。结果表明slit3在HT1080细胞中的不表达可能与启动子的甲基化有关,外源性的slit3可显著抑制HT1080细胞的增殖和侵袭转移的能力。小鼠静脉注射实验进一步验证了slit3对于HT1080细胞迁移和转移的抑制作用。

xon guidance molecules,slits are secreted proteins that play an important role in axonal guidance, branching and neuronal migration. In recent years,slits are also found to involve the regulation of tumor growth,angiogenesis and metastasis. Fibrosarcoma,HT1080 cell ,which derived from mesenchymal tissues, is a highly metastatic malignant tumor.In this study,the expresion of slit3 and its effects on proliferation,migration and invasion of HT1080 are examined. The results showed that the silence of slit3 expression in HT1080 may be associated with promoter methylation.Addition of ectogenic slit3 suppressed the proliferation,migration and invasion of HT1080 significantly,which was further confirmed in vivo by intravenous injection experiment.
