苎麻叶片再生及根癌农杆菌介导的遗传转化参数研究

Using leaves derived from test-tube plantlets as explants in ramie tissue culture and Agrobacterium-mediated transformation can maintain the biological characters of the original plants. Leaves explants derived from test-tube plantlets of Huazhu No. 4 and Huazhu No. 5 were used to evaluate the effects of different media on plant regeneration in this study, in which the highest regeneration ratio was up to 50%. The optimum medium for plant regeneration from Huazhu No. 4 was MSB medium supplemented with 0.5 mg/L thidiazuron (TDZ) and 0.01 mg/L IAA, and the best one for Huazhu No. 5 was MS medium containing 0.5 mg/L TDZ and 0.01 mg/L NAA. In the Agrobacterium-mediated transformation of ramie, the optimal cocultivation duration, acetosyringone (AS) concentration and Agrobacterium concentration was 72h, 50-100 mg/L and OD6000.5- 0.8, respectively, which will be a practical tool for the ramie transformation in the near future.
