【目的】从转移相关基因方面研究γ-生育三烯酚对胃腺癌SGC-7901细胞转移的影响。【方法】本实验采用体外细胞培养胃腺癌SGC-7901细胞,分别用0、15、30、45、60μmol/L的γ-生育三烯酚对其进行处理48小时,再运用RT-PCR技术对胃腺癌转移相关基因的mRNA进行检测。【结果与结论】1.γ-生育三烯酚抑制SGC-7901细胞基质金属蛋白酶MMP-2、MMP-9的表达;2.γ-生育三烯酚提高SGC-7901组织细胞基质金属蛋白酶抑制剂TIMP-1、TIMP-2的表达;3. γ-生育三烯酚增加抑制肿瘤转移基因nm23-H1的表达。

Objective Study the effects of gamma-tocotrienol on the genes related with metastasis in human gastric madenoccarcinoma SGC-7901 cells. Methods First, we cultured SGC-7901 cells and then treated with γ-T3 at doses of 0、15、30、45、60μmol/L for 48 hours and then measured mRNA expression of genes related to the metastasis of SGC-7901 cells by RT-PCR. Results and Conclusions 1. γ-tocotrienol inhibited the expression of matrix metalloproteinase in SGC-7901 cells; 2. γ-tocotrienol improved the expression of tissue inhibitor of matrix metalloproteinase in SGC-7901 cells; 3. γ-tocotrienol improved the gene nm23-H1 expression in SGC-7901 cells.
