目的 研究原花色素对口腔常见致病菌和生物膜的影响,探讨其抗菌防龋机制。方法 通过体外抗菌实验,测定原花色素对口腔常见致病菌的最小抑菌浓度,观察变异链球菌在原花色素作用下的生长情况,动态监测原花色素对变异链球菌产酸的影响,并进一步研究原花色素对变异链球菌生物膜生长代谢的影响。结果 ① 原花色素对变异链球菌,格氏链球菌的最低抑菌浓度为4mg/ml,对聚核梭杆菌及牙龈卟啉单胞菌的最低抑菌浓度分别为1mg/ml和0.25mg/ml;② 原花色素在MIC浓度下能够有效抑制变异链球菌产酸;③ 原花色素可以抑制变异链球菌生物膜的形成,MBIC50为0.0625 mg/ml,0.5mg/ml原花色素可抑制50%的生物膜代谢活性。结论 原花色素在不影响其他测试细菌生长的情况下,可以抑制变异链球菌生长、产酸和生物膜的形成,并抑制已经形成的生物膜的代谢活性。

Objective To evaluate the effect of PA on the oral bacteria and on the biofilm formation and metabolism Methods The minimum inhibitory concentrations (MIC) were determined using the microdilution method. The effect of PA on the acid production of oral bacteria were evaluated at a sub-MIC concentration. The biofilm formation and metabolism were determined by crystal violet stain and XTT method. Results ① PA inhibited the growth of tested oral bacteria. The M IC of PA on S. mutans and S.gordonii was 4m g /m l , on F.nucleatum and P.gingivalis was 1mg/ml and 0.25mg/ml ; ② PA can inhibit the acid production of S. mutans signifigently ; ③ it was also proved that the growth and the metabolism of biofilm formed by S. mu tans, would be inhibited under the effect of PA .Conclusion Proanthocyanidin effectively infected the cariogenicity of oral bacteria and biofilm, Which may be a promising natural agent for caries therapy.
