热致形状记忆性人工晶状体材料的研究

Study on Thermally Induced Shape-memory Intraocular Lens Materials

【目的】研究交联剂的分子量和含量对新型热致形状记忆性人工晶状体材料性能的影响。【方法】利用弯曲试验在模拟体内环境下对材料的形状记忆性能进行表征。利用接触角测量仪测量材料的浸润性。与小鼠成纤维细胞系L929细胞共培养7天,CCK-8法检测材料细胞毒性。与人晶状体上皮细胞系SRA01/04共培养24小时,结晶紫染色法检测材料表面晶状体上皮细胞黏附。与荧光标记的纤维连接蛋白培养2小时后荧光显微镜下观察表面蛋白吸附。用细胞培养小室系统研究晶状体上皮细胞在材料下面的移行。【结果】八种试验材料均能在40秒内完全恢复至初始形状,且无明显细胞毒性。交联剂分子量为400的四种材料的LEC黏附无显著差异,纤维连接蛋白的吸附较多,LEC在材料下面的移行受阻;而交联剂分子量为1000的材料,随着交联剂含量的增加,其黏附性能降低,LEC移行增多。故交联剂分子量为400的材料的囊膜相容性较好。

Objective] Study the effects of different molecular weights and concentrations of the crosslinking agents on the new thermally induced shape-memory intraocular lens materials. [Methods] Use bending test to detect the shape-memory properties in simulated vivo condition. Use contact angle measuring instrument to detect the infiltration. Measure the cytotoxicity with CCK-8 assay. Use the crystal violet assay to detect lens epithelial cell adhesion on the material surface. Observe the surface protein adsorption under fluorescent microscope after 2 hours cultured with fluorescently labeled fibronectin. Research the lens epithelial cell migration below the material with cell culture chamber. [Results] Eight test materials can fully restore to its original shape within 40 seconds, and show no significant cytotoxicity. The materials of crosslinking molecular weight 400, with more LEC adhesion, fibronectin adsorption and higher adhesion characteristics, LEC migration below the material are blocked. However, the materials of crosslinker molecular weight 1000, with the crosslinking agent concentration increases, the adhesion characteristics decrease and LEC migration increase. Therefore, the materials of crosslinking molecular weight 400 show better capsular biocompatibility.
