乳腺癌是目前常见的女性的恶性肿瘤,其发生、演进、侵袭和转移是一个多基因参入的渐进积累的过程,涉及到许多基因调控、表达、突变等变化。人的SLP-2基因是一个新的肿瘤相关基因,在多种肿瘤的发生过程中发挥重要作用,特别是其在乳腺癌组织的高表达与病人的存活率相关。因此为了进一步研究SLP-2在乳腺癌中的功能,应用免疫印记的方法检测SLP-2在不同乳腺癌细胞中的蛋白表达水平,发现随着细胞癌化程度的增强其表达水平也增强,并且成功构建pCMV-Myc-SLP-2重组质粒,证明了其在MCF-7细胞的表达,MTT实验结果表明SiRNA转染乳腺癌细胞MCF-7可以抑制细胞增殖,同时我们也发现SLP-2在乳腺癌细胞系中表达量与HER2/nue呈负相关。这些结果都为深入研究乳腺癌的发生发展奠定了基础。

Breast cancer is locally advanced at the time of diagnosis,and its occurrence, evolution, invasion and metastasis is a gradual accumulation process of multi-genes participating, it also involves gene regulation, expression, mutation and many other changes. Human stomatin like protein 2 (SLP-2) is a novel cancer-related gene,it played important roles in a variety of tumors, especially high-level SLP-2 expression were associated with significantly decreased survival in breast cancer.To study the function of SLP-2 in breast cancer furtherly,we detected the expression of SLP-2 protein in different breast cancer cells by Western blotting,and found that the expression of its protein in breast cancer cells increased with the increase of the degree of cancer.We also successfully constructed recombinant plasmid pCMV-Myc-SLP-2 and demonstrated it can expressed well in MCF-7. The MTT assay showed that the cell proliferation was significantly inhibited after transfection of pCMV-Myc-SLP-2, we also found that the expression of SLP-2 and HER2/nue have negative correlation in MCF-7, thus providing a useful clue for further research on the occurring and development of breast cancer.
