以赤霞珠(Vitis vinifera L. cv. Cabernet Sauvignon)葡萄果实为试材,研究了8℃低温锻炼对葡萄果实DFR变化的影响以及在高温45℃胁迫下的适应性反应;研究了水杨酸对DFR在酶活、蛋白及转录水平的影响。结果显示,低温锻炼初期,DFR在mRNA、蛋白以及活性水平相继被激活;经过低温锻炼后的葡萄果实在高温45℃胁迫下,DFR活性在各水平均比对照高,表明低温锻炼提高了果实对高温胁迫的适应性。水杨酸处理果实DFR在mRNA、蛋白以及活性水平相继激活。以上结果表明,温度和水杨酸调控了DFR在mRNA、蛋白以及活性水平的代谢,并且这个调控反应是快速和敏感的。

Effects of low temperature (8℃) acclimation and high temperature (45℃) stress on dihydroflavonol 4-reductase (DFR) were investigated in grape berries (Vitis vinifera L. cv. Cabernet Sauvignon)and the impact of salicylic acid (SA) on the gene expression of DFR at enzymic activity, translational and transcriptional levels was also analyzed. The results showed that the expression of the gene encoding DFR increased at mRNA, protein and activity levels, respectively, at the early period of low temperature acclimation. The gene expression at various levels was present more significant increase in the low temperature-acclimated grape berries during heat stress than that in the non-low temperature acclimated berries, which indicated that low temperature acclimation could promote the adaption of grape berries to heat stress. SA pretreatment also activated the gene expression of DFR at various levels. It was thus suggested that both temperature acclimation and SA were involved in the regulation of DFR gene expression, and the regulation was rapid and sensitive.
