丝状真菌(Filamentous fungi)的发酵生产通常具有较高的工业应用价值,但其菌体形态是一个区别于其他非丝状菌的一个重要指标。有鉴于此,本研究创新性地建立了一套完整的自动化形态分析方法,能够在短时间内获得大量真实可靠的菌体形态参数,大大减小图形分析过程中投入的劳力。同时,本文还应用此方法,研究了在黑曲霉发酵生产糖化酶过程中不同供氧水平及剪切水平对菌体形态的影响。通过大量形态数据定量阐明了黑曲霉形态受供氧及剪切水平影响的规律,为进一步工业过程优化提供了重要基础数据。

Filamentous fungi is widely applied in industrial fermentation. Moreover, the particular fungi morphology acts as a fatal index for a successful fermentation. Here, a systematical morphology analysis procedure was developed to obtain abundant morphological information efficiently. Meanwhile, this methodology was successfully applied to the study of Aspergillus niger morphology under different oxygen supply conditions and shear rate conditions. As a result, the responding pattern of Aspergillus niger morphology under these conditions was quantitatively demonstrated, which laid a solid foundation for the scale-up of fungal fermentation.
