本研究以玉米BAC片段为探针,在玉米根尖有丝分裂的细胞学制片上进行BAC-FISH研究,分别探讨了玉米和水稻基因组C0t DNA对探针重复序列的封阻、杂交后洗脱的严谨度、杂交液中FAD的浓度变化等因素对BAC-FISH定位的影响,进一步优化了玉米BAC探针在玉米根尖有丝分裂染色体上进行FISH研究的技术体系。

he four randomly picked BAC-DNA were used as probes and Fluorescence in situ Hybridization (FISH) has been applied to interphase and metaphase mitotic chromosomes. For optimizing BAC-FISH technique using on maize, we have respectively assayed the effect on some factors, such as maize or rice genomic C0t DNA used as blocking DNA, washing temperature and FAD concentration in washing buffer and in hybrid solution. As a result, a set of optimized BAC-FISH parameter which may is available to maize mitotic chromosomes was proposed.
