基因工程菌自诱导IL-7蛋白表达的研究
Study of protein expression of IL-7 of engineering E. coli through auto-induction ways
本工作采用乳糖自诱导的诱导培养替代传统异丙基硫代乳糖苷(IPTG)的诱导,研究乳糖自诱导培养菌种的生长和蛋白的诱导表达规律;对乳糖浓度、接种量和溶解氧浓度对乳糖自诱导培养的影响亦进行了研究。实验结果表明,利用乳糖自诱导,在不添加抗生素的条件下,可以达到与IPTG类似的诱导效果,菌体浓度可达到IPTG的诱导水平的171%。
he possibility of using lactose auto-induction to substitute the common inducer IPTG in the fermentation process of the recombinant microorganism in E.coli was deeply investigated. The influences of culture conditions such as lactose concentration, inoculums concentration, DO concentration on the expression of the recombinant protein were analyzed and studied in detail. The result indicated that lactose could induce foreign protein expression and enhance cell growth during the induction period. The IL-7 expression level of auto-induction without antibiotics in medium could got as same the level as that with IPTG as inducer. And the OD600 reached about 171% of the level with IPTG as inducer.
