肝癌血清中岩藻糖结合蛋白的分离与鉴定

Isolation and Identification of Fucose-binding Proteins in Sera of Hepatocellular Carcinoma

目的] 分离并鉴定肝细胞癌血清中的岩藻糖结合蛋白,并为新的癌症诊断标志物的发现提供新思路。[方法] 利用岩藻糖功能化的磁性微粒进行岩藻糖结合蛋白的分离,并利用LC-MS/MS 进行鉴定。[结果] 健康人血清中分离并鉴定岩藻糖结合蛋白49种,肝癌患者血清中分离并鉴定岩藻糖结合蛋白45种,其中,健康人与肝癌患者血清同时出现的蛋白为26种。通过非标记定量方法emPAI定量比较,有8种蛋白在肝癌中低表达,3种高表达。[结论] 本研究分离并鉴定了健康人和肝癌患者血清中的岩藻糖结合蛋白,同时,肝癌中高表达的糖结合蛋白可以作为肝癌诊断的潜在标志物。

urrent biomarkers used clinically still suffer from poor sensitivity and specificity in the diagnosis of hepatocellular carcinoma (HCC). Therefore, new strategy is still needed for discovery of biomarker in HCC. Methods: Here a strategy was performed for the isolation and identification of fucose-binding proteins from the clinical sera (60 normal donors and 60 hepatocellular carcinoma patients) by the fucose-magnetic particle conjugates-based method and LC-MS/MS analysis. Results: 75 proteins for normal donors and 79 proteins for HCC cases were identified, of which 26 were identified in both samples. With the exponentially modified Protein Abundance Index (emPAI) quantification, 8 fucose-binding proteins were estimated down-regulated (emPAI ratio > 2) and 3 MBPs were estimated up-regulated (emPAI ratio < 0.25) in the HCC sera. Conclusions: Fucose-binding proteins were identified as up-regulated in the HCC could be served as potential biomarker and this research also provided new strategy for the discovery of new biomarkers.
