镉对Zn/Cd超富集植物滇苦菜(Picris divaricata Vant.)氮素代谢的影响
Effect of Cadmium on Nitrogen Metabolism of Zn/Cd hyperaccumulator Picris divaricata Vant.
以采用水培的方法,研究了不同镉水平(0、5、10、25、50、75μM)对超富集植物滇苦菜叶片中硝态氮、铵态氮、游离脯氨酸、可溶性蛋白质含量以及硝酸还原酶、谷氨酰胺合成酶、谷氨酰胺转化酶、谷氨酰胺脱氢酶活性的影响。结果表明,镉处理降低了滇苦菜的生物量,25μM~75μM处理对其有明显的毒害作用。镉处理还降低了硝酸还原酶、谷氨酰胺转换酶的活性,但对谷氨酰胺脱氢酶的活性影响不大,对谷氨酰胺转换酶的活性在一定程度上有促进作用。随着镉处理浓度的增高,硝态氮、铵态氮的变化趋势是先降低后升高,分别在50μM处理和25μM处理时达到最低,并没有出现积累的情况。游离脯氨酸含量在高处理的情况下升高的很明显,缓解了高浓度镉处理对滇苦菜的毒害作用。
Hydroponic culture was conducted to study the effect of Cd on the growth , metal accumulation and nitrogen metabolism in Picris divaricata Vant. The enzymatic activities of nitrogen metabolism including nitrate reductase (NR) , glutamine synthetase (GS) , GS-transferase(GOGTA) and glutamate dehydrogenase(GDH),as well as free proline , soluble protein , NO3--N and NH4+-N in Picris divaricata Vant were determined.Result indicated that the addition of Cd redused the biomass and high level of Cd(25μM~75μM) could significantly poison Picris divaricata Vant.Compared to the control plants,Cd treatments decreased the activites of NR,GS but increased the activites of GOGAT,however ,neither of NO3--N nor NH4+-N were cumulate.Both the contents of NO3--N and NH4+-N decreased with the Cd concentration. The contents of NO3--N becomed the lowest when level of Cd is 50μM,as well as NH4+-N inμmoloL-1 treatment. The increase of free proline might alleviate the toxicity of high level of Cd in Picris divaricata Vant.
