基于表型的棉花遗传多样性分析及核心材料的筛选

在DNA多样性与表型多样性还难以对应的现阶段,基于表型的遗传多样性分析和核心材料的筛选是育种工作和基础研究工作的前提和基础。本文通过对来自三个栽培种(陆地棉、 海岛棉和亚洲棉)的92份棉花资源的9个性状的调查和分析,变异系数、特异材料%和Shannon-Weaver多样性值均表明该群体蕴含较丰富的遗传变异;利用软件NTSYS-pc,采用类平均法(UPGMA, unweighted pair group method arithmetic averages)对相似系数矩阵和遗传距离矩阵进行聚类分析,结果表明品种间的遗传关系与品种所属的种及系谱吻合性较高;利用软件QGAStation筛选出核心材料22份,相关统计学指标(方差差异百分率、均值差异百分率、极差符合率和变异系数变化率)显示核心材料组遗传多样性与原群体基本一致。因此,可以认为基于表型的遗传多样性分析及核心材料的筛选将为后续的育种工作和基础研究提供重要的参考信息。

In that DNA diversity detected nowadays could not mean phenotypic diversity, it is the precondition of breeding project and basic research of crop improvement that genetic diversity analysis and sampling of core collection by phenotypes. Ninety-two cultivars (belonging to G. hirsutum L, G. barbadence L or G. arboreum L) were phenotyped on nine traits, and the statistic (variation coefficient, special p.c., Shannon-Weaver diversity index, etc.) indicated that there exists high level of phenotypic diversity of 9 traits among the 92 accessions. According to the software NTSYS-pc, the cluster analysis by the unweighted pair group method of arithmetic (UPGMA) means were conducted based on the pair similarity and genetic distance, and 92 accessions were classified. The classification is mostly consistent with species and known pedigree information. Using the software QGAStation, 22 accessions were sampled from the original population to make up the core collection. The statistic (the mean difference percentage (MD%), the variance difference percentage (VD%), the coincidence rate (CR%) and the variable rate (VR%)) of the core collection in terms of the original population showed that the genetic diversity of core collection was approximately equal to the original population. The results of genetic diversity analysis and sampling of core collection by phenotypes would provide reference for many projects (e.g. parent-selection of hybridization, genetic-material-selection of basic research).
