目的:观察INS-1细胞中CASK基因对微丝肌动蛋白(F-actin)的作用对胰岛素分泌过程的影响。方法:1. 体外培养大鼠INS-1细胞,明确CASK对于高钾刺激的胰岛素分泌功能(KSIS)的影响。2.使用免疫荧光法观察CASK对F-actin分布和形态的影响。3.使用流式细胞仪观察CASK对F-actin含量的影响。结果:1. 在INS-1细胞中,干扰CASK的表达能够造成细胞KSIS功能损伤(P<0.01)。2. CASK同F-actin可能共同在以细胞膜附近区域协同调节胰岛素的分泌,CASK能够造成F-acitn局限性聚集分布。3. CASK的干扰或者过表达均不能引起F-actin的含量显著变化。结论:1. CASK的表达降低与INS-1细胞KSIS功能减退相关。2. CASK能够造成F-acitn局限性聚集,从而使得其分布局限化,减弱了其阻碍锚定分泌的屏障作用,从而促进了胰岛素的分泌,这可能是CASK调节胰岛素囊泡的分泌的机制之一。3. CASK对F-actin含量没有明显影响。

Objective: Investigate the influence of insulin secretion process by CASK effect on microfilaments F-actin. Methods: 1.Cultured INS-1 cells, certified the impact of CASK on the K+ stimulate insulin secretion (KSIS)function in INS-1 cells . 2.To detect the impact of CASK to the distribution and form of F-actin by using immunofluorescence. 3.To investigate whether CASK change F-actin content by using flow cytometry. Results: 1. Interference the level of CASK was able to reduce KSIS function in INS-1 cells. 3.Both CASK and F-actin might be coordinately regulated insulin secretion in the cell membrane, CASK caused local assembly of F-acitn in INS-1cells. 3. The interference or overexpression of CASK could not change content of F-actin. Conclusion: 1. CASK is associated with KSIS function in INS-1 cells. 2. CASK induce local assembly of F-actin which result in the distribution of F-actin is localized, accordingly,it reduces the barrier role of F-actin obstruct anchor process in insulin secretion ,hence ,it is a possible mechanism that insulin vesicle secretion regulated by CASK. 3. CASK can not affect the content of F-actin.
