野菊花组织培养与离体快繁技术研究
Study on the Tissue Culture and Rapid Propagation of Chrysanthemum indicum
以野菊花为材料,通过对其离体叶片的组织培养,建立了诱导、分化、增殖、生根等一整套离体快繁体系。结果表明,野菊花叶片外植体较好的灭菌方法为70%酒精5s+2% NACLO 8min;叶片外植体初代培养的适宜培养基为MS+ 0.2 mg.L-1 6-BA + 0.2 mg.L-1 NAA,培养温度25℃,培养时间30d,光照12h/d;继代及增殖培养以MS+ 0.2 mg.L-1 6-BA + 0.1 mg.L-1 NAA配比时效果较好,培养温度25℃,培养时间30d,光照12h/d;试管苗生根培养的最适宜培养基为1/2MS基本培养基附加0.2 mg.L-1 NAA,培养温度25℃,培养时间30d,光照12h/d。
In this paper, with the leaves of Chrysanthemum indicum as explants, a set of propagation system in vitro about the tissue culture of induction, differentiation, proliferation, rooting was established. The results showed that the optimal sterilization method for leaves of Chrysanthemum indicum was 70% alcohol 5s + 0.2% NaClO 8 min; optimal early culture medium was MS+ 0.2mg.L-1 6-BA + 0.2mg.L-1 NAA, the temperature is 25℃,the incubation time is 30d and illumination is 12h/d in this method, on which the leave explants could grow vigorously;the optimal proliferation culture medium among all tested media was MS+ 0.2mg.L-1 6-BA + 0.1mg.L-1 NAA. The temperature is 25℃,the incubation time is 30d and illumination is 12h/d;the optimal rooting culture medium among all tested ones was 1/2MS basic medium added 0.2 mgoL-1 NAA. And the temperature is 25℃,the incubation time is 30d and illumination is 12h/d.
