柳珊瑚内生真菌Aspergillus versicolor的天然产物研究
Natural products from a gorgonian-derived fungus Aspergillus versicolor
目的:揭示海洋柳珊瑚内生真菌Aspergillus versicolor代谢产物的结构特征与结构多样性;方法:采用大米固体培养技术对微生物进行实验室规模培养,采用现代色谱技术分离和纯化化合物,采用现代波谱技术确定化合物的结构;结果:从海洋真菌Aspergillus versicolor中获得23种天然产物,并确定了它们的化学结构,其中化合物1为新颖结构,确定了文献尚未确定的化合物2的绝对构型,补充归属了化合物2的碳谱数据,发现化合物1-3具有抗肿瘤瘤谱作用;结论:研究结果表明不同来源的同一微生物物种产生的天然产物结构多样性不同,提示同一菌株在不同生态条件下所激活的生物合成通路不同,并表明同一菌株生物合成途径多样,不同环境产生的突变体激活不同合成途径。
Objective: to uncover the typical structures and the structural diversity derived from a gorgonian endophytic fungus; Method: rice solid fermentation was used for the fungus culture in laboratory scale, comprehensive column chromatography was applied for the isolation and purification of compounds, extensive spectroscopic data were used for the structure determination; Results: 23 natural products were isolated, and their structures were determined, including a new compound (1), while the absolute configurations of compound (2) which were uncertain in the literature were assigned in present work, compounds 1-3 exhibited inhibitory effects against a panel of tumor cell lines;Conclusion: Chemical examination of the marine-derived fungus A. versicolor resulted in the isolation of 23 natural products, including a new compounds. The absolute configurations and the NMR data of compound (2) which were uncertain in the literature were assigned in the present for the first time. The inhibitory effects compounds 1-3 against a panel of tumor cell lines suggested 1-3 to be the model molecules for further structural modification. The present work indicated that the same strain of fungus encoded diverse biogenetic pathway, while a fungal strain from different location activated synthetic pathway to produce distinct secondary metabolites.
