目的 研究脐带来源的间充质干细胞(mesenchymal stem cells,MSCs)对同种异体外周血单个核细胞(peripheral blood mononuclear cell, PBMC)来源的γδ T细胞主要生物学功能如细胞因子分泌及细胞毒作用的影响。方法 MSC与活化的PBMC(IL-2和PAM刺激12天)共培养48小时(比例MSC:PBMC=1:5,1:80),流式细胞术分析γδ T细胞功能性细胞因子(IFNγ,TNF, perforin, IL-10,Granzyme B)分泌情况;同样MSC与活化的PBMC共培养60小时,后磁珠分离γδ T细胞,纯化的γδ T细胞(E)对感染流感病毒H1N1的A549细胞(T)以E:T=10:1比例进行杀伤,流式细胞术分析杀伤结果。结果 实验结果显示MSCs对γδ T细胞生物学功能有抑制作用, MSCs明显抑制γδ T细胞IFNγ的分泌(p<0.05),TNF, perforin, IL-10的表达无明显变化(p>0.05),但Granzyme B的产生有升高趋势(p<0.05);MSCs可使γδ T细胞杀伤感染了流感病毒H1N1的A549细胞的作用明显抑制(p<0.05)。结论 MSCs对γδ T细胞的细胞因子分泌及细胞毒作用均有明显的抑制作用。

In this paper, a serial of experiments were designed to determine biological changes in cytokine secretion and cytotoxicity of γδ T cells after co-cultured with UC-MSCs in vitro. MSC were cocultured with PBMCs stimulated by IL-2 and PAM for 12 days at a ratio of 1:5 or 1:80 for 48 hours, and secretion of functional factors such as IFN gamma, TNF, perforin, IL - 10, Granzyme B ofγδ T cells were analyzed by flowcytometry. After the same coculture system for 60 hours, γδ T cells were purified to cocultured with influenza virus H1N1 infected A549s at E:T=10:1,and the dead A549s were analyzed by flowcytometry. The biological function of γδ T cells was inhibited by MSCs. The secretion of IFN gamma was obviously inhibited by MSCs (p < 0.05), and there was no significant change in expression of TNF, Perforin and IL-10 (p>0.05), but the generation of Granzyme B has a rising trend (p < 0.05); but the generation of Granzyme B has a rising trend (p < 0.05). The cytotoxicity of γδ T cells against influenza virus infected A549 was significantly inhibited by MSCs (p < 0.05).
