慢性应激对大鼠淋巴细胞DNA损伤机制的研究

目的] 研究慢性应激对大鼠淋巴细胞DNA损伤机制。[方法] 通过建立慢性应激动物模型,测定动物血清皮质醇的含量;电镜观察海马超微结构改变,黄嘌啉氧化酶法测定SOD活性,TBA法测定MDA含量,彗星实验评价大鼠外周血淋巴细胞DNA损伤情况。[结果] 实验组海马超微结构异常;血清SOD活性(56.70±15.65 )U/ml降低, MDA含量(9.21±2.75)nmol/ml升高(P﹤0.05),SOD活性与皮质醇负相关(P﹤0.05),MDA含量与皮质醇正相关(P﹤0.05),彗星细胞发生率56.5%(P﹤0.05);实验组淋巴细胞DNA损伤率增加(P﹤0.05);DNA损伤与MDA含量正相关( P﹤0.05),。[结论] 慢性应激可损伤海马超微结构,慢性应激可诱导机体产生过量的自由基,促进脂质过氧化损伤外周血淋巴细胞DNA。
Objective] To study DNA damage in peripheral blood lymphocytes under chronic stress. [Methods]By established chronic stress model, and then measured levels of cortisol in blood.Observed the pathomorphism changes in hippocampus with electron microscope. And then the activities of serum superoxide dismutase (SOD) was measured with xanthine oxidase method and the contents of malondiasldehyde (MDA) with thiobarbituric acid in two groups. The degree of DNA damage in peripheral blood lymphocytes was analyzed with comet assay. [Results] Observed the pathomorphism changes in hippocampus with electron microscope. The activities of SOD were lower(56.70±15.65 )U/ml,and contents of MDA in stressful group were higher than the control(9.21±2.75)nmol/ml(P﹤0.05).The activity of SOD were lower(P﹤0.05),and contents of MDA were higher(P﹤0.05) in stressful group.The rate of DNA damage was higher in peripheral blood lymphocytes of rats in stressful group than control(P﹤0.05). It was positive correlation among serum MDA level. [Conclusion] Chronic stress damage hippocampus,Chronic stress may increase contents of MDA and decrease activities of SOD, give rised to some pathomorphism changes in spleen and DNA damage in peripheral blood lymphocytes of rat.
