Identification of ASH2 gene and mRNA expressionunder heat stress in Plutella xylostella

小菜蛾Plutella xylostella(Linnaeus)是十字花科植物的主要害虫之一,ASH2在翅发育过程中具有重要作用,但在昆虫中此类基因极少被报道。本文采用PCR和RACE技术克隆了小菜蛾Plutella xylostella翅基因ASH2。克隆结果表明,ASH2基因核苷酸序列全长809 bp,编码231个氨基酸,估测其成熟蛋白分子量2.55kD,等电点为9.02。Blast结果表明与家蚕(Bombyx mori L.)ASH2基因的同源性达到85.75%。运用RT-qPCR技术研究不同温度(25℃-8h、40℃-8h;25℃-8h、42℃-8h;25℃-1h、44℃-1h)处理后小菜蛾蛹的表达量。结果表明热激处理导致小菜蛾蛹ASH2基因表达量明显下降,推测高温下小菜蛾翅发育受损可能与该高温胁迫下该基因表达受抑有关。

iamondback moth (DBM), Plutella xylostella, is one of destructive pest of cruciferous crops. ASH2 gene plays a central role in wing development, but little work has been reported on this gene in insects. In this research, ASH2 was cloned by applying PCR and RACE technologies.The results showed that the ASH2 gene in diamondback moth, named ASH2(Gen Bank accession no. KJ158243), is 809 bp in length and encodes 231 amino acid residues. The results of blast showed that cloned gene has 85.75% nucleotide sequences homology with Bombyx mori . The expression of pupa was studied by applying RT-qPCR at different temperature (25℃-8h、40℃-8h;25℃-8h、42℃-8h;25℃-1h、44℃-1h). The results indicated that the expression level of ASH2 gene was reduced sharply after heat shock treatment. It speculated that the DBM wing development was damaged at high temperature which is related to the expression of ASH2 gene was reduced at the high temperature.
