目的 了解猪苓多糖(PPS)及猪苓多糖联合卡介苗(BCG)体内刺激对小鼠腹腔巨噬细胞TLRs表达的影响。方法 应用流式细胞术检测PPS联合BCG体内刺激2小时、12小时、24小时后小鼠腹腔巨噬细胞表面CD14,TLR2,TLR4及黏附分子CD11b的表达。结果 PPS组在作用的3个时点内小鼠腹腔巨噬细胞TLR分子表达无明显变化,联合BCG作用2小时后TLR表达高于BCG和PPS单用组;PPS组作用12小时后,CD11b表达高于对照组,24小时后下降,联合BCG作用2小时和12小时,CD11b表达高于BCG和PPS单用组。结论 PPS体内单独刺激不能促进TLR分子的表达,联合BCG可促进TLR分子和黏附分子CD11b的表达。

Objective:To learn the effect of injection with PPS and/or BCG on the expression of TLRs on mouse peritioneal macrophage. Method:Flow cytometry was used to measure the expression of CD14, TLR2, TLR4 and CD11b on mouse peritoneal macrophage 2h, 12h, 24h respectively after injection with PPS and/or BCG. Results:In PPS group, expression of TLRs on mouse peritoneal macrophage did not make significant difference amoung 3 timepoints, while combined with BCG induced higher expression of TLRs at 2h timepoint, compared to injection with BCG or PPS alone. Injection with PPS induced higher expression of CD11b than that with salt at 12h timepoint, then went down at 24h timepoint. Combined with BCG induced higher expression of CD11b than that with BCG or PPS alone at 2h and 12h timepoints. Conclusion: Injection with PPS alone dose not enhance the expression of TLRs on mouse peritoneal macrophage, while combined with BCG would induce higher expression of TLRs and CD11b.
