MYB21 is a key transcrition factor in jasmonate signaling to coordinate late stamen development in Arabidopsis thaliana. The promoter activities of MYB21 and At3g27809, a gene produces a natural antisense RNA of MYB21, were investigated by promoter fusion to GUS gene. Moreover, we generated the GFP fusion protein to test the cellular location of MYB21 and At3g27809. The main results are as followed: MYB21 promoter activity is detected in stamen filament, stigma, vascular tissue of sepal and petal as well, and is strongly induced by methyl jasmonate; At3g27809 promoter is active in the stamen filament vascular tissue, stigma and is not induced by methyl jasmonate; Diven by the conjugated promoter of MYB21 andAt3g27809, GUS expression is similar to that driven by MYB21 pomoter, and is strongly induced by methyl jasmonate in flower organ; MYB21-GFP fusion protein was exclusively localized in nuclear, and At3g27809-GFP fusion proteins was mainly localized in nuclear as well. The present study also suggests that At3g27809 may play a role in regulating the expression of MYB21. Further study will shed light on the regulatory mechanism of spatial and temperal expression of MYB21.
