苯酚降解菌的分离及其降解效率的测定

从郑州市郊农田和池塘等土样中,用富集培养的方法筛选出能高效降解苯酚的菌株DP-1、DP-5、DP-16。实验表明,DP-5的降解能力较好,在培养基中苯酚含量为500mg/L时,其降解效率可达68%;苯酚含量为1500mg/L 时,降解效率为56%。苯酚含量测定采用4-氨基安替比林吸光光度法,各试剂的加入顺序对结果影响较大。葡萄糖的存在对苯酚的降解没有促进效果,相反,会由于对葡萄糖的优先利用,苯酚的降解能力大幅下降。
hree objective strains, named as DP-1、DP-5 and DP-16 respectively, were selected from the samples, which were taken from farm and ponds in the suburbs of Zhengzhou, by methods of enrichment medium culture, so that the strains could live on phenol as sole carbon source. It was showed that DP-5 had the best abilities of degradation in comprison with the other two strains, and when the concentrations of phenol were 500mg/L and 1500 mg/L, the degradation efficiency could be up to 68% and 56% respectively. The content of phenol could be determined by 4-Amino Antipyrine Spectrophotometry, but the results could be affected greatly by the adding order of reagents. The presence of glucose did not promote its degradation efficiency expectedly, on the contrary, because of the priority in the use of glucose, the degradation capacity dropped significantly.
