脐带来源的间充质干细胞的分离、传代培养及其分化为成骨和成脂细胞
Isolation, culture, osteogenic and adipogenic differentiation of human umbilical cord-derived mesenchymal stem cells
目的:探讨从人脐带中分离和培养脐带间充质干细胞的方法,并通过诱导分化实验研究其多向分化的能力。方法:通过组织块植入法分离人脐带间充质干细胞;将脐带间充质干细胞诱导分化为成脂和成骨细胞。结果:组织块植入法培养7天后,有少量细胞从组织块中游离出来,呈射线状排列,细胞为梭形;传代培养的细胞成涡旋或平行排列生长,增值较快,每2~3天需传代;成脂诱导分化21天后,油红O可将脂肪滴染成亮红色;成骨诱导分化21天后,茜素红可将钙结节染成红色。结论:组织块植入法是一种分离脐带间充质干细胞的较好方法,可以将脐带间充质干细胞诱导分化为成骨和成脂细胞,表明脐带间充质干细胞具有多向分化的潜能。
Objective: Explore the method for isolation and culture of human umbilical cord-derived mesenchymal stem cells(hUC-MSCs) from the umbilical cord, and study the potential for the multiple differentiation of hUC-MSCs. Methods: hUC-MSCs were isolated form human umbilical cord by tissue implant method; hUC-MSCs were induced to the osteogenic and adipogenic differentiation. Results:After 7 days of culture, a few cells migrated out from the placenta tissue and grew in ray-like arrangement, Cells were passaged every 2 to 3 days and grew parallelly; After adipogenic differentiation for 21 days, fat droplets can be stained bright red by oil red O; After osteogenic differentiation for 21 days, calcium nodules can be stained red by Alizarin red. Conclusion:Tissue implant is a good method to obtain hUC-MSCs. hUC-MSCs can be induced to differentiate into osteogenic and adipogenic cells, indicating that hUC-MSCs have the potential of multi- differentiation.
