目的:对盐酸苯肼诱发网织红细胞的实验条件进行优化,对诱发时所用的盐酸苯肼剂量、诱发时间、动物的品系等条件进行比较。 方法:ICR小鼠及BALB/c小鼠,每天背部皮下注射1%盐酸苯肼100μl~400μl,连续注射9d. 结果:注射400μl、300μl剂量盐酸苯肼的小鼠诱发的网织红细胞不健康,且很快死亡,注射200μl、100μl剂量盐酸苯肼的小鼠诱发的网织红细胞较为健康,最高比例出现在第4d,可达50%~70%. 第6d后,血液中网织红细胞比例下降,红细胞增多。结论:ICR小鼠及BALB/c小鼠注射最佳剂量和时间为每只每天注射200μl,连续注射4~5d。ICR小鼠对盐酸苯肼的耐受性明显强于BALB/c小鼠,但二者诱发出的网织红细胞无明显差异。

Objective. To optimize the best conditions for induced mouse reticulocyte,we compared with the different conditions such as dose of injection, time and species of mouse. Methods. The 1% hytrazinobenzene hydrochloride was subcutaneously injected to the back of the ICR and BALB/c mouse everyday. The doses were 100μl, 200μl, 300μl, 400μl, respectively for 9 days. Results. The morphology of reticulocytes from the mouse that injected 400μl and 300μl hytrazinobenzene hydrochloride was abnormal, and the mouse was dead in few days, but injected 200μl or 100μl hytrazinobenzene hydrochloride was better than others. The ratio of induced reticulocyte is about 50%~70% (day 4), but the ratio of erythrocyte is increased after the sixth day. Conclusion. The best conditions for induced reticulocyte are to inject 200μl per mouse everyday for 4 or 5 days. The ICR mouse showed that was more tolerance for hytrazinobenzene hydrochloride than BALB/c mouse.
