环糊精葡萄糖基转移酶(Cyclodextrin Glycosyltransferase,简称CGTase,EC 2.4.1.19)是α-淀粉酶家族(家族13)中的主要成员。它作为一种多功能酶,可催化四种不同的反应,其中通过环化反应可以将淀粉及其衍生物转化为环糊精(CD)。本研究将来源于Bacillus circulans 251的β-CGTase经过密码子优化后,通过亚克隆在Escherichia coli BL21(DE3) 中表达,构建基因工程菌E. coli BL21(DE3)/pET-20b(+)-β-CGTase。摇瓶发酵中,重组菌发酵48 h后胞外酶活达20 U/mL。
yclodextrin glycosyltransferase (CGTase, EC 2.4.1.19) is the member of α-amylase family (family of 13). As a multi-functional enzyme, it can catalyze four different transglycosylation reactions. CGTase is capable of converting starch or starch derivates into cyclodextrins (CDs) through cyclization reaction. In the present study, the optimized gene of β-CGTase from Bacillus circulans 251 was subcloned into E. coli BL21(DE3). In shake flask , the extracellular enzyme activity was 20 U?ml-1 after cultivation of 48 h.
