【目的】为了探索肾素基因启动子甲基化水平与精神分裂症可能的相关性及潜在的生物学作用。【方法】利用外周血基因组,采用Massarray飞行时间质谱法,测定肾素基因启动子甲基化水平。采用剑桥神经心理自动化成套测试中的快速视觉信息处理Rapid Visual Information Processing, RVP)和延迟匹配(Delayed Matching To Sample, DMS)评估受试者注意力及视觉记忆。【结果】发现两组间肾素基因启动子甲基化水平无显著差异。以性别、年龄、患病状态做协变量,肾素基因启动子甲基化水平与RVP和DMS一些指标间具有显著相关。【结论】肾素基因可能通过启动子甲基化在精神分裂症病理中发挥重要的作用。

o explore the potential role of renin gene promoter methylation on pathogenesis of schizophrenia, we tested the methylation level of CpG in renin gene promoter from peripheral blood of schizophrenia patients and healthy controls. The rapid visual information processing (RVP) and delayed matching to samples (DMS) tests of Cambridge Neuropsychological Test Automatic Battery were performed to assess the Association between renin gene methylation and schizophrenia cognitionattention and visual memory of subjects form Association between renin gene methylation and cognitive impairments in schizophreniatwo groups. The association between CpG methylation levels of renin gene promoter and the two cognition domains were analyzed by partial correlation. The methylation levels of all CpG in renin gene promoter showed no significant difference between two groups. After setting gender, age and disease status as covariate, the methylation level of CpG cite chr1:204166158 (hg38) as significantly correlated with RVP total correct rejection (r=-0.68, p=2.42×10-7), RVP A\' (r=-0.61, p=6.94×10-6), DMS total correct(r=-0.55,p=1.02×10-4), DMS total correct all delays (r=-0.54, p=.52×10-4). The renin gene may play an important role in the pathology of schizophrenia through promoter methylation.
