家蚕Bax inhibitor-1基因的原核表达及功能分析
Expression and Functional Analysis of the Bax inhibitor-1 Protein from Silkworm Pupa (Bombyx mori)
对本实验室构建的家蚕cDNA文库进行筛查时,发现一条与Bax inhibator-1有较高同源性的基因,生物信息学分析后,初步命名为BmBI-1(家蚕Bax inhibator-1),GenBank登录号是DN237509。为初步研究BmBI-1的功能,将该基因克隆到表达载体pET-28a(+)中,重组成原核表达质粒pET-28a(+)-BmBI-1,转化感受态细胞E.coli BL21(DE3), IPTG诱导表达并纯化该重组蛋白,制备多克隆抗体。用家蚕Bm5细胞进行亚细胞定位研究表明BmBI-1主要分布于细胞质中,可能定位于内质网等细胞器上。同时用荧光定量PCR和western blot的方法分析不同组织及发育时期该基因的表达差异,结果显示在各组织中BmBI-1表达量从高到底依次是马氏管、睾丸、头、肠、气门、表皮、脂肪体、丝腺;在卵、幼虫、蛹和蛾四个发育时期中,蛾中的表达量最高,其次是幼虫,再次是蛹,而在卵中几乎没有检测到该蛋白。从以上试验结果可以看出,BmBI-1可能与其他已知类似蛋白相同,在家蚕的发育和维持正常生理功能中起重要作用。
When screening Silkworm cDNA library, we found a gene which has a higher homology with Bax inhibator-1 and named BmBI-1 (Bombyx mori Bax inhibator-1). In order to research the function of BmBI-1, the ORF of this gene was cloned into the pET-28a(+) vector, and constructed the procaryotic expression plasmid pET-28a(+)-BmBI-1. The recombinant plasmid was transformed into E.coli BL21 (DE3). After inducing with IPTG , the analysis of SDS-PAGE showed that the recombinant protein was expressed compared to control. The recombinant protein was soluble and was purified with metal-chelating affinity chromatography. Immunization of New Zealand rabbit with recombinant protein generated high titer polyclonal antibodies (1:12800). Subcellular localization study with polyclonal antibodies in silkworm cell Bm5 suggest that BmBI-1 is mainly distributed in the cytoplasm, may be positions in the endoplasmic reticulum.At the same time, The gene expression features in different organizations and differences stages were analysis using fluorescence quantitative PCR and western blot method. The results showed that in all tissues BmBI-1 are expressed as following from the most to the lest : Malpighian tubes, the testicles, head, intestines, valves, skin, fat body, silk gland; In the four Development stages, moth the is the highest followed by larva, and the next is pupa ,but in eggs , few expression was detected. From the above results we can have a conclusion that BmBI-1 may be related to development and maintenance of normal physiological functions in silkworm as other similar proteins.
