丹参雄性不育与可育近等基因系中EST-SSR差异标记的筛选
Screening of EST-derived microsatellites in near-isogenic lines of male sterile and fertile Salvia miltiorrhiza
利用雄性不育是实现作物杂种优势利用最经济、最有效的途径之一。为寻找与丹参雄性不育基因紧密连锁的EST-SSR标记,对从GenBank上下载的鼠尾草属11747条EST序列进行处理,在获得高质量EST序列后,经MISA软件搜索和分析EST序列中SSR位点的分布规律。运用Oligo 7软件设计并合成76对EST-SSR引物,以不育/可育近等基因系(NIL)为对象,筛选出10对在不育和可育NIL基因池中表现出差异。将这10对引物在两NIL群体中进行再次验证,发现仅有SmSSR56和SSR19两对引物分别在可育和不育NIL群体中表现出差异,与丹参中育性调控基因紧密连锁。
Heterosis is one of the economic and the most effective approaches to improve crop yield. To identify EST-derived microsatellite (EST-SSR) markers linked to the sterile gene in S. miltiorrhiza, a total of 11,747 EST sequences included in Salvia genus were downloaded from GenBank for screening. The high quality ones selected from these EST sequences were analyzed to seek SSR sites after processing. On the basis of sequence analysis, 76 primer pairs were synthesized by using Oligo 7 software, and then were screened by using sterile and fertile near-isogenic lines (NIL) gene pools as example. Ten primer pairs were found to have different fragments in both gene pools, and then were confirmed in sterile and fertile NIL populations. Two primer pairs, SmSSR56 and SSR19, exhibited different fragments in fertile and sterile NIL populations, respectively, which indicate both are tightly linked to fertility-related gene in S. miltiorrhiza.
