补阳还五汤对局灶性脑缺血大鼠脑内caveolin的影响
Effects of Buyanghuanwu Decoction on expression of caveolin in a Rat Model of Focal Ischemia
目的:探讨补阳还五汤对局灶性脑缺血大鼠脑内caveolin的影响。方法:采用大脑中动脉阻塞法复制脑缺血模型,动物随机分为正常组、假手术组、模型组、补阳还五汤组,给予不同处理,分别于12h,1,3,7天处死动物,采用免疫组织化学法观察caveolin1、2的表达。结果:补阳还五汤能显著减轻神经功能缺损;正常脑内有一定量的caveolin1、2表达,缺血后caveolin1、2迅速增加,分别于术后第3或1天达高峰,补阳还五汤组显著提高caveolin1、2的表达,在第3、7天模型组与补阳还五汤组差异有显著性(p <0.01)。结论:补阳还五汤可调节脑缺血后caveolin1、2的表达,是其促进神经功能恢复的可能机制之一。
Objective:To investigate the effect of BYHW decoction on expression of caveolin in a rat model of cerebral ischemia. Methods: Brain ischemia was induced by middle cerebral artery occlusion. The rats were randomized into four groups including naive, shame-operated, ischemia, and ischemia treated with BuYangHuanWu Decoction (BYHW). The BYHW groups of rats were then orally administered daily with the BYHW and killed at 12h,1, 3 or 7day point after operation. Caveolin1 and caveolin2 were measured by immunohistochemical staining. Results: There were some Caveolin 1 and 2 positive cells in normal brain , expression of caveolin 1 or 2 was enhanced immediately after ischemia and reached peak at 3 day or 1 day after ischemia respectively ; BYHW decoction increased the number of caveolin1-positive or caveolin2-positive cells. This increase was statistically significant at 3 and 7 day after ischemia and treatment. Conclusion: BYHW decoction is capable of regulating expression of caveolin 1 and 2 in ischemic brain , that may be an anti-ischemic mechanism of BYHW decoction.
