花色苷是酿酒红葡萄果实中重要的水溶性色素类物质,通常只积累在成熟红葡萄果实的果皮之中。然而对于"染色"葡萄,花色苷则会同时大量积累于其果皮和果肉。本研究旨在探索欧亚种染色葡萄品种'Tintorera'转录因子VvMYBA1基因的启动子在其果肉花色苷生物合成组织特异性中的潜在功能与作用,通过分子生物学技术克隆其DNA序列,通过生物信息学对其潜在功能进行预测,通过启动子重组载体侵染烟草叶片来检验其基本功能。研究发现,染色葡萄品种'Tintorera'VvMYBA1基因的启动子序列与已报道的'黑比诺'VvMYBA1的启动子序列相同,都具有多种光响应元件,以及其它转录因子和不同激素信号的响应位点,因而不是染色葡萄果肉中花色苷组织特异性积累产生的根本原因。此外,该基因启动子活性检验表明,这些目标序列具有启动子的基本活性,其重组载体在烟草叶片中能够启动报告基因的大量表达。

nthocyanins are important water soluble flavonoid pigments in winemaking red grapes, which were only accumulated in the skins of the matured red grapes normally. But for the "teinturier" grapes, anthocyanins could accumulate greatly in both of their skins and pulps. The present study was aim to investigate the potential functions and effects of the key transcription factor VvMYBA1 in the anthocyanin tissue-specific biosynthesis of its pulps. Therefore, its DNA sequence was cloned by using molecular biology technology, its potential functions was predicted with the help of bioinformatics, and its basic functions were tested by using infecting of promoter combinant vector on tobacco leaves. This research revealed that the promoteSequence Cloning and Functional Analysis for the Promoter of the Key Transcription Factor VvMYBA1 in Anthocyanin Biosynthesis in the Teinturier grape Vitis Vinifera L. cv. 'Tintorera'r sequence of VvMYBA1 of the teinturier grape cultivar 'Tintorera' was the same with the reported one from 'Pinot Noir', which had various photoresponse elements, as well as the response sites to other different regulators or phytohormone signals. Besides, the function test revealed that the VvMYBA1 promoter had the basic biological functions of promoters, the combinant vector of which could start the great expression of the report gene on tobacco leaves.
