脑内注射不同浓度Aβ25-35拟阿尔茨海默病模型在Morris水迷宫行为学测试中学习记忆的差异
Learning and Memory Differences in Morris Water Maze Behavioral Tests of Different Alzheimer's Disease Model Prepared by Intracerebral Injection with Different Concentrations of Aβ25-35
目的:观察不同浓度的Aβ25-35侧脑室注射后大鼠Morris水迷宫学习记忆能力的变化,探讨注射制备AD模型大鼠时Aβ25-35的最佳浓度。方法:雄性SD大鼠随机分为假手术组和模型组,模型组注射Aβ25-35浓度分别为2μg/μl、4μg/μl和8μg/μl。参照《大鼠脑立体定位图谱》,选取右侧侧脑室注射聚集态的Aβ25- 35,制备AD大鼠模型。造模成功后7天采用Morris水迷宫测试各组大鼠学习记忆能力的变化。结果:逃避潜伏期结果显示,与假手术组比较,模型组大鼠逃避潜伏期时间明显增加,差异均有统计学意义(P<0.05);与模型组中2μg/μl组大鼠比较,4μg/μll组与8μg/μl组逃避潜伏期时间明显增加,差异有统计学意义(P<0.05);4μg/μl组与8μg/μl`组间比较,差异无统计学意义(P>0.05)。目标象限的活动时间与路程显示,与假手术组比较,模型组注射不同剂量Aβ25-35的大鼠目标象限活动时间和路程均显著减少,差异具有统计学意义(P<0.05),但模型组不同浓度间比较差异无统计学意义(P>0.05)。空间探索结果显示,与假手术组比较,模型组注射不同剂量Aβ25-35的大鼠穿越平台次数均显著减少,差异具有统计学意义(P<0.05),与模型组中2μg/μl组大鼠比较,4μg/μl和8μg/μl组穿台次数明显减少,差异具有统计学意义(P<0.05)。4μg/μl组与8μg/μl组间比较,差异无统计学意义(P>0.05)。结论:单侧侧脑室注射Aβ25-35制备大鼠AD模型时,注射Aβ25-35的推荐浓度为4μg/μl。
bstract Objective: To investigate the changes of Morris water maze learning and memory ability of AD model rats after injected by different concentration of Aβ25-35 into lateral ventricle, and to discuss the optimal concentration of Aβ25-35 to prepare AD rats. Methods: Male SD rats were randomly divided into sham operation group and model group, and rats in model group were injected with Aβ25-35, with concentrations of 2μg/μl, 4μg/μl, and 8μg/μl. According to the Rat Brain Stereotaxic Atlas, aggregation of Aβ25-35 was injected into the right side of the lateral ventricle to prepare AD rat model. 7 days after successful modeling, Morris water maze was applied to test rats learning and memory ability. Results: The results showed that compared with the sham group, the escape latency time of rats in model group significantly increased, and the differences were statistically significant (P<0.05); compared with the model group of 2μg/μl, the escape latency time in 4μg/μl group and 8μg/μl group significantly increased, and the difference was statistically significant (P<0.05); between 4μg/μl group and 8μg/μl group, the difference was not statistically significant (P>0.05). Active time and distance of target quadrant showed that compared with the sham group, rats of model group injected with different doses Aβ25-35 significantly reduced, and the difference was statistically significant (P<0.05), but the difference between different concentrations in model group was not statistically significant (P>0.05). Space exploration results showed that compared with sham-operated group, platform-crossing time of rats in model group injected with different doses of Aβ25-35 were significantly reduced, and the difference was statistically significant (P<0.05). Compared with the model group of 2μg/μl, the platform-crossing time in 4μg/μl group and 8μg/μll group significantly reduced, and the difference was statistically significant (P<0.05). And between 4μg/μl group and 8μg/μl group, the difference was not statistically significant (P>0.05). Conclusion: the recommended concentration of Aβ25-35 to injected into unilateral ventricle to prepare AD model was 4μg/μl.)
