紫草素(Shikonin)与阿卡宁(Alkannin)是一对手性对映体,具有广泛、显著的生物活性。许多研究表明从紫草属(Boraginaceae)中制备紫草素或阿卡宁的对映体过量存在明显的差异,然而目前定性定量测定其对映体纯度的旋光光度法和液相色谱法存在诸多不足。本文首次在辽宁硬紫草素侧链上引入S-(-)-α-甲氧基-α-(三氟甲基)-苯乙酸形成Mosher酯,并运用1HNMR测定后者H-1' 化学位移的非等价性所产生的峰值相对强度,从而确定辽宁硬紫草素对映体纯度,其结果准确性被手性液相色谱法得以验证。

Shikonin and alkannin, a pair of chiral enantiomer, have been found to demonstrate a wide range of significant biological activity. Many studies have also shown that the enantiomeric excess (e.e.) of shikonin or alkannin from the roots of boraginaceae varied significantly, however, the current qualitative and quantitative determination of the e.e. mainly came to the aid of the optical spectrophotometry and liquid chromatography, which there are many disadvantages to influence their accuracy. In the paper, we first introduced S-(-)-α- methoxy-α- (trifluoromethyl)- phenyl -acetic acid to hydroxyl group in the side chain of shikonin isolated form Lithospermum erythrorhizon Sieb. et Zucc to form Mosher esters, and then used 1HNMR to determine the relative intensity of the resulting peak of H-1' as a result of chemical shift non-equivalence, and thereby to acquire the e.e of shikonin prepared, and finally the accuracy of e.e. was to verify by chiral liquid chromatography.
