果糖诱导高尿酸血症大鼠肾脏转运蛋白Glut9研究
Research of Renal Transporter Glut9 on Hyperuricemia in Rats Induced by Fructose
目的:从肾脏转运蛋白角度探究果糖诱导的高尿酸血症大鼠的病理机制。方法:将20只雄性SD大鼠按体质量随机分为2组,正常组给予清水,模型组给予10%果糖饮水建立高尿酸血症模型,2组均给予正常饲料,实验第40天处死。检测大鼠血清尿酸(SUA)、尿尿酸(UUA)水平,计算肾脏尿酸清除率(CUA),并测定肝脏黄嘌呤氧化酶(XOD)活性。利用免疫组化观察Glut9在各组大鼠肾脏的表达位点并计算其蛋白表达含量,逆转录实时荧光定量PCR(RT-qPCR)检测各组大鼠肾脏转运蛋白Glut9 mRNA表达水平。结果:与正常组比较,实验第10d-40d,模型组大鼠血尿酸(SUA)水平显著升高,实验期间尿酸清除率无显著性差异。与正常组相比,实验第40d模型组大鼠肝脏黄嘌呤氧化酶(XOD)活性无显著性差异,其肾脏Glut9蛋白及基因表达水平均上调。结论:10%果糖饮水可成功诱导大鼠高尿酸血症模型;果糖诱导高尿酸血症病理机制可能与上调肾脏Glut9蛋白及基因表达水平,增加肾脏尿酸重吸收相关。
IM: To explore the pathological mechanism of hyperuricemia rats induced by fructose from the aspects of renal transporter Glut9. METHODS: Twenty SD male rats were randomly divided into two groups by weight. The normal group was given water, the model group was given 10% fructose water to establish hyperuricemia model. At the same time, all groups were fed with normal feed. The rats were sacrificed on the 40th day. We detected serum uric acid(SUA) and urinary uric acid(UUA) to calculate the clearance rate of uric acid in renal(CUA). The activity of hepatic xanthine oxidase(XOD) was need to be tested. Meanwhile, we examined the expression level of renal transporter Glut9 mRNA using real-time quantitative reverse transcription-polymerase chain reaction (RT-qPCR) method. RESULTS: From the 10th to the 40th experimental days, the SUA of the model group raised significantly than the normal group, but the CUA and XOD activity of renal had no difference. On the 40th day, the expression level of protein and gene of Glut9 in the renal of model group were higher than that in the normal group. CONCLUSION: Fructose drinking can induce hyperuricemia in rats, which is probably related to the up-regulation of the expression level of protein and gene of renal Glut9 ,and the increase of reabsorption of renal.
