银-石墨烯纳米复合物标记电化学适配体传感器用于血清中凝血酶的检测
Ultrasensitive electrochemical aptasensor for thrombin detection using silver-decorated graphene nanosheets as labels
本文构建了一种新型的电化学适配体传感器用于血清中凝血酶的检测。首先,在玻碳电极(GCE)表面电化学沉积一层纳米金颗粒,进而化学吸附氨基化的凝血酶适配体,从而制得高灵敏电流型适配体传感器。以甲醛作为还原剂、聚乙烯吡咯烷酮为表面活性剂,将纳米银修饰到石墨烯的表面,用于凝血酶适配体的标记。在凝血酶存在下,具有氧化还原特性的纳米复合物就会交联到传感器的表面,从而导致电化学信号的产生。在优化的实验条件下,该适配体传感器的响应电流与凝血酶浓度的对数呈现线性关系,线性范围为0.01pM - 1.0 nM,检出限为3.33 fM。
In this paper, a new electrochemical aptasensor was designed for determination of thrombin using silver-decorated graphene nanosheets as labels on aptamer/nanogold-modifed glassy carbon electrode. Growth of silver nanocrystals on graphene was prepared based on one-step method by using formaldehyde as reducing agent and poly(N-vinyl-2-pyrrolidone) as the surface modifier under mild conditions. The as-prepared silver-graphene nanocomposites were empolyed for the labelling of aminated aptamers. The assy was carried out in PBS, pH 7.4 with a sandwich-type assay mode by using the decorated nanosilver as indicators. Under optimal conditions, the electrochemical aptasensors exhibited a wide linear range from 0.01 pM to 1.0 nM thrombin with a low detection limit (LOD) of 3.33 fM. The precision, selectivity and stability of the aptasensors were acceptable. Importantly, the methodology was monitored with thrombin spiked sample in blank fetal calf serum, and the recovery was 91 - 123%.
